Immunochemical techniques practical Abstract Neat solutions of antigen (BSA) proved to give stronger interactions with the antiserum resulting in the greater formation of antigen : antibody complexes. Also different antibodies/antigens diffuse differently to produce the equivalence point due to the mass and charge difference of these molecules present. Resulted on the formation of different precipitation points around the antiserum. Studies on the technique ‘ouchterlony double diffusion’, with firstly dilutions of HSA as the antigen and antiserum to HSA raised in goats. Secondly using antigens of unknown mixtures of IgG and/or BSA with the antiserum to BSA and IgG. The results from experiment 1 showed that there was one complete line around the antiserum between the six wells. This line however got weaker in appearance as the solution of BSA got weaker.
