...dure: A 5g sample of hamburger meat and 10mL of buffer was obtained, placed in a large centrifuge tube, and mixed thoroughly. The buffer solution was use to release the myoglobin from the meat. It was centrifuged for 15 minutes to make sure the myoglobin was separated from the meat. The supernatant was taken from the centrifuge tube and placed in a cuvet. The absorbance was read on the supernatant at different wavelengths with the spectrophotometer. Then the supernatant was divided into two cuvets half into one with potassium ferricyanide half added to one cuvet containing supernatant and sodium dithionite and then the absorbencies were obtained.
